This is a TIRFM image of the basal surface of a mouse myoblast expressing the PH domains of myosin 10, fused to GFP. The image was generated using the TIRF microscope built by Gregory Mashanov and Justin Molloy at NIMR, Mill Hill London. The image is colour coded - highest levels of PH123-GFP are in red, and lowest in Blue. Spots on the membrane that correspond to single molecules of PH123-GFP can be tracked to measure their appearance and disappearance at the membrane, and their lateral mobility. |
